| I. Background | |
IL-1 家族包括 11 種細(xì)胞因子，7 種具有激動劑活性的配體(IL-1α， IL-1β，IL-18, IL-33, IL-36α， IL-36β， IL-36γ)和 4 種具有拮抗活性的成員[IL-1 受體拮抗劑(IL-1Ra)， IL-36Ra, IL-37, IL-38]。IL-1α， IL-1β， IL-18, IL-33, IL-36α， IL-36β， IL-36γ觸發(fā) MPKK 和 NF-κB 導(dǎo)致炎癥反應(yīng)。而 IL-1 家族的其他成員具有抗炎作用。IL-1 是該家族的一個標(biāo)志性細(xì)胞因 子，它可以共同刺激 T 輔助細(xì)胞、B 細(xì)胞的成熟和增殖、NK 細(xì)胞的激活，并與炎癥有關(guān)。 IL-1 由上皮細(xì)胞、內(nèi)皮細(xì)胞、血小板、成纖維細(xì)胞和免疫細(xì)胞(如巨噬細(xì)胞和中性粒細(xì)胞) 產(chǎn)生和分泌，作為先天和適應(yīng)性免疫反應(yīng)的一部分，保護機體免受損傷和感染。 IL1α或 IL1β結(jié)合 IL1R1 誘導(dǎo) IL1R1 與 IL1R 輔助蛋白(IL-1RAcP)二聚化，由此產(chǎn)生的 復(fù)合體招募 MyD88，進而招募 IL-1 受體相關(guān)激酶 1 (IRAK1)。IRAK1 激活 TNF 受體相關(guān) 因子 6 (TRAF6)/ TGFβ活化激酶(TAK1)/ κ b 激酶信號級聯(lián)，導(dǎo)致核因子κB (NF-κB)抑制劑 i -κB α的磷酸化和降解，從而釋放細(xì)胞質(zhì) NF-κB 轉(zhuǎn)運到細(xì)胞核，激活炎癥和免疫基因的轉(zhuǎn)錄。 | |
