I. Background | |
抗體依賴的細(xì)胞介導(dǎo)的細(xì)胞毒性作用(ADCC,antibody-dependent cell-mediated cytotoxicity )是指抗體的 Fab 段結(jié)合病毒感染的細(xì)胞或腫瘤細(xì)胞的抗原表位,其 Fc 段與殺 傷細(xì)胞(NK 細(xì)胞、巨噬細(xì)胞等)表面的 FcR 結(jié)合,介導(dǎo)殺傷細(xì)胞直接殺傷靶細(xì)胞,它是 抗腫瘤的治療性抗體藥物發(fā)生作用的一種作用重要機(jī)制。 傳統(tǒng)用于測定 ADCC/ADCP 的方法主要依賴于相關(guān)原代免疫細(xì)胞的分離、體外分化, 進(jìn)而測量靶細(xì)胞的殺傷或吞噬效應(yīng)。這些方法高度依賴供體原代細(xì)胞,價格昂貴且費(fèi)時費(fèi) 力,對原代細(xì)胞的分離分化過程對實驗操作的要求非常高,實驗失敗風(fēng)險較大,由于分離 分化細(xì)胞純度問題,還存在檢測信號低以及不穩(wěn)定均一的問題。另外,每次實驗的細(xì)胞供體來源可能都不一致,實驗結(jié)果可重復(fù)性較差,可能存在批次 差異。由于以上這些因素的影響,在需要高質(zhì)量控制的藥物開發(fā)過程中,很難建立穩(wěn)定可 靠的檢測方法。 | |
